Abstract:

Background: Infertility affects 15% of couples around the world. Male factor accounts for 40-50% of conjugal infertility. cAMP responsive element modulator (CREM) is a key factor in the regulation of the expression of number of postmeiotic genes that control spermatogenesis. Aloe vera is a medicinal plant used to treat several diseases. The purpose of this study was to investigate effects of Aloe vera extract on rat spermatogenesis with emphasis on sperm parameters, hormonal assay, and CREM gene expression in testis tissue. Materials and Methods: Aloe vera leaf pulp extractgel extract, and a mixture of both administered to three groups of 10-week old male Wistar rats for 56 consecutive days. Then sperm analysis, hormonal assay, and RT-PCR were carried out. Results: Results indicated that in all groups except control group the weights of the testes have increased. Epididymal sperm counts and sperm motility have been increased significantly compared to control groups. Analysis of testosterone level between groups showed that the level of this hormone in the groups that treated with Aloe Vera has increased remarkably. Results of RTPCR showed an increase in the CREM gene expression in the groups that received gel extract and mixture of gel and leaf pulp extract. Conclusion: According to the results of this study, Aloe Vera has strong spermatogenic activity by increasing sperm parameters and CREM gene expression. This study strongly proposes that Aloe vera specially its gel fortifies spermatogenesis and can be a good candidate for manufacturing fertility drugs.

P-37: (+)-Pulegone Reduced Cyp19 GeneExpression and Reduced Leydig CellsSteroidogenic Activity

Background: Pulegone (PGN) is a clear colorless oily liquid, which is widely used in different areas for flavoring foods, drinks, and dental products. There are several reports indicating that the metabolites of the PGN affect cytochrome-P450 family expression and activities. The cytochromes-P450 (cyp19) are involved in Leydig Cells aromatization. Therefore, present study was performed in order...

P-85: Epididymal Sperm Parameters following Sperm Mediated Gene Transfer of EDFP-pIRES Vector in Rat

Background: Sperm mediated gene transfer was approved for producing transgenic animals. There are many studies which reported different success rate by this procedure. Failure in this project mostly is returned to transfection and stability of transferred genes. The other problem that may contribute in project failure may return to the testicular damages which occur at the time of transfection,...

P-47: Comparison of Sperm Acrosin Activity and DNA Fragmentation between Varicocele and Infertile Individuals

Background: Introduction assessment of sperm functional tests along with routine semen analysis may provide further information in the etiology of varicocele in different subpopulations. Therefore, in this study, in addition to assessment of sperm chromatin integrity, we aimed to assess the total acrosin activity which has proven to have major role during in vitro and natural fertilization in v...

P-61: Effects of Acrylamid on Sperm Parameters and Chromatin Quality in Mice

Background: Acrylamid (AA) is found in carbohydraterich foods that are prepared at high temperatures, such as French fries and potato chips. The toxic effects of AA in male animals include degeneration of epithelial cells of seminiferous tubules, decreased number of sperm and elevation in abnormal spermatozoa. The aim of this study was to evaluate the detrimental effects of AA on sperm paramete...

P-12: The Improvement of Sperm Parameters and Chromatin Quality by Vitamin E Supplement in Mice

Background: Vitamin E is believed to be the primary component of the antioxidant system in spermatozoa. There is a relationship between activity of these antioxidant and function of sperm. Vitamins E and C which are belong to non-enzymatic antioxidant are used as a supplemented drug to improve sperm quality and male fertility. The present study was carried out to investigate the positive effect...
